Cultivate Mycobacterium species with Thermo Scientific™ Remel™ Dubos Polysorbate 80 Albumin Broth.

In 1946, Dubos and Davis described a liquid medium in which growth of tubercle bacillus was obtained from dilute inocula in 3 to 5 days1. Dubos and Middlebrook2 then studied the use of this medium to cultivate Mycobacterium tuberculosis. The original solution was modified in 1950 by Dubos, Fenner and Pierce3 by adding a strong buffering system and an acid pH which provided dispersed growth from a small inoculum. This broth is used for subculturing mycobacteria and preparing inoculum for other in vitro tests4.

Dubos Polysorbate 80 Albumin Broth is a liquid medium recommended for cultivation of Mycobacterium species. Polysorbate 80 is an oleic acid ester and an in vitro indicator of mycobacterial virulence. Virulent cultures form cords in media containing oleic acid or its esters, while nonvirulent cultures grow more diffusely.

Dubos Polysorbate Albumin Broth contains casein peptone and asparagine that supply nitrogen for the growth of mycobacteria. Disodium and monopotassium phosphates provide the buffering system. The inorganic salts supply ions essential for metabolism. Polysorbate 80 (10%) serves as a nutritional source of essential fatty acids and also acts as a dispersing agent for homogenous growth from a small inoculum. Bovine plasma albumin (serum factor V) enhance the growth of mycobacteria and provides protection against bacteriostatic and bactericidal effect by toxic agents.
